Confocal microscopy uses a laser light source to excite fluorescent probes within cells, thereby generating fluorescence images of subcellular fine structures inside cells or tissues. Building on the principles of wide-field microscopy, the laser scanning confocal microscope uses a laser beam as its light source. If the sample contains fluorophores that can be excited, the emitted fluorescence travels back along the original excitation path, re‑entering the beamsplitter. In the optical design, only light from the focal plane is allowed to pass through the detection pinhole; light originating from regions outside the focal plane is blocked. Consequently, the background at non‑focused locations appears dark.
